Honors Bridge Club certainly claims to be the largest in the US and Jeff Bayone has used this as part of product marketing but I do not know if there is any official data supporting the claim.

Clubs in the US usually don't have membership in the same way that European clubs often do. There isn't a yearly or monthly fee, and anyone can show up to play in a game (often without more than a few minutes advance notice). They do charge people to play of course, and there may be an additional charge for people who are not ACBL members (but that membership is at the zonal level). People who are ACBL members have a "home unit" but this is just based on their address and has nothing to do with paying any fees to a club or even playing in the clubs (many of the better players just play in tournaments and not clubs) -- and units sometimes include multiple clubs.

For this reason there isn't a clear answer to the question. Clubs do maintain table counts over the year (Honors in NY has the most for a face-to-face club, although of course BBO has a lot more) but this will count players multiple times (if I play once a week, I've added 13 total tables to the yearly count all by myself) and can also count guests who would not normally be considered club members.

Around here, some clubs do have membership. Anyone can play, but members get extra benefits.

For example, at one local club, paying the annual membership fee lets you play in unlimited games instead of paying per game, but you also occasionally have to help with the setup/teardown (the playing site is a church assembly room). At another club, membership reduces your entry fees (you break even on your third game) and makes you eligible for free plays when you win. Members also get to vote on club policies.

What do you mean by "club"? In the widest sense, the biggest US club is the ACBL.
