Playing mini roman 4-4-4-1 or 4-4-5-0 10-12 pts with 4 spades if opponents interfere over 2♦ what should a double by openers partner mean?
Take out or penalty? I am thinking that if opener has shortnest in the suit he should bid or if it is one of his 4 card suits he should pass?

As one of the few pairs who actually use R2DII, but range of 11-14, any singleton, no void...we have some views which might be applicable to your conditions:

After a simple overcall, a double is not stiff or void in the O/C suit. It is willing to defend right there if pard has 4 of them, but able to play in another strain if pard has a stiff.

A pass commits opener to balance with a double if short.

I don't know how the spade guarantee should affect this. As a side note, Phil posted a thread with 1-4-4-4 and 12-14 HCP as a rebid problem which we don't have and one of the reasons we use the 2D toy ---to get the dreaded minimum 4X1's out of our other auctions, not to jack the opponents around.
