Do you have the queen of trumps ? In an auction that has gone (spades agreed) 4N(blackwood)-5♦(say 1 or 4)-5♥ an explanation of ?Q would be sensible.
There are other relay and asking sequences where you might ask for the Q of a specific suit.

Typically, the affirmative is to reply as if partner had asked for Kings (or Specific King) and the negative is to return to trumps.
But it can be different, for example we play that 5N within a control-bid sequence denies Queen of trumps and skipping 5N to show a control affirms.
